CRISPR screen analysis: a buyer’s review checklist

A nonclinical scoping checklist for CRISPR screen provenance, data QC, hit robustness, confounders and independent follow-up evidence.

An existing CRISPR screen can generate a useful development lead without validating a target. The source map identifies independent screen interpretation as a specialist service. This checklist defines the information needed to scope that review; it is not a guide to designing or executing an editing experiment.

Identify the screen before interpreting it

Record the library and version, sample structure, intended comparison, metadata and original analysis. A final ranked table may be insufficient to review the result. Ask which questions can be answered from available files and which would require additional authorized data.

Make robustness a deliverable

Request a documented explanation of which findings persist under the agreed checks and which depend on a particular analysis choice. Require exclusions and deviations to be visible. A reviewer should state when the data cannot support the proposed comparison instead of returning a confident ranking regardless.

Keep confounders separate from target biology

The commercial aim is to determine what the evidence justifies next. A high-ranked hit is not automatically a causal therapeutic mechanism. Alternative explanations and the need for independent validation belong in the report. Qualified scientific review is a prerequisite, not an optional marketing claim.

Ask for a reproducible handover

Agree the input specification, versioned code or notebook, review outputs and decision memo. Confirm permissions before data move. The public inquiry form is for a nonconfidential description, not research files or patient information.

Treat commercial rights as a separate workstream

The Broad Institute’s licensing guidance distinguishes noncommercial academic work from commercial uses. Buying a reagent does not necessarily authorize every downstream commercial activity. The analysis scope, later laboratory work and licensing questions each need their own review. Patent counsel—not a screen ranking—resolves legal rights questions.

The practical takeaway: Buy a traceable interpretation and a justified next question, not an unsupported promise of target validation.
