Research by Gizem Gumuskaya and coauthors, including Michael Levin. Karam’s contribution here is an explicit evidence audit and research workflow.
Cells maintain different ion concentrations across their membranes. Selective channels permit ionic currents; pumps use energy to maintain gradients. Membrane potential reflects this coupled physical system.
The explorer approximates this with capacitance, leak conductance and a fixed reversal potential. It does not simulate pumps or changing concentrations.
Gap junctions provide direct communication between adjacent cells. Electrical coupling can distribute a local perturbation through connected cells. Connectivity and conductance influence the response.
Try the zero-coupling preset: changing a neighbor can no longer affect an isolated cell through this modeled connection.
Cell shape, adhesion, contact and tissue forces change the context in which cells communicate. A morphological shift may also reflect changes in cell composition or selection. These explanations need separate measurements.
Mechanics is a research question in K02. No mechanical state or morphology prediction is computed by the passive network explorer.
Electrical state, signaling, gene regulation and physical organization can interact through feedback. A causal explanation requires suitable perturbations and measurements that distinguish competing mechanisms.
A younger molecular-clock estimate is one observation. Durable recovery of a useful function is a separate endpoint.
Original explanatory diagrams, not microscopy or experimental data. Framework informed by the credited primary sources.
Cells maintain electrical differences across their membranes. Ion channels provide selective conductance; pumps use energy to sustain ion gradients. Gap junctions can couple neighboring cells, so a local perturbation may alter a larger network. Membrane potential interacts with biochemical signaling, transport and cell behavior; it is one part of the system.
Mechanics belongs in the same question. Geometry, contact, adhesion and tissue forces influence which cells communicate and how a construct behaves. Feedback can connect voltage, channel activity, gene regulation and physical organization. Observing a pattern does not identify which feedback loop caused it.
The framework is informed by Michael Levin and collaborators’ published work, including Gizem Gumuskaya’s Anthrobot research, Alexis Pietak’s modeling, Hamid Sediqi’s keratinocyte work and Nirosha Murugan and colleagues’ regeneration study. Author and source register. No Tufts partnership, endorsement or ownership of their discoveries is implied.
Which electrical variable changes which cellular process? Perturbation and orthogonal measurements are needed to distinguish cause from correlation.
How do local interactions produce a stable shape or collective behavior? Geometry and cell selection can change outcomes without an age-reversal mechanism.
Does a system recover useful function after damage? Gap coverage, regrowth and sensorimotor outcomes answer different questions.
Does a clock track functional improvement in the same biological context? Preserve donor hierarchy, clock identity, handling and composition.
A model makes assumptions explicit and generates conditional predictions. It does not validate them in living tissue.
Human-derived cells are not treated human participants. None of the five sources establishes a human rejuvenation therapy or universal switch.
| Condition | Clock estimate (years) | Reported n |
|---|---|---|
| Passage-1 baseline | 23.7 | 12 |
| Anthrobot day 10 | 19.1 | 6 |
| Anthrobot day 25 | 20.0 | 3 |
| Air–liquid interface | 25.7 | 3 |
Figure 5 reports 23.7 → 19.1 years. The main text instead describes 25 → 18.7 and approximately 25%. These are conflicting source statements, not interchangeable estimates. Reported n values are not donor counts.
Clock algorithms/version, preprocessing and biological/technical replicate hierarchy remain UNKNOWN: the supporting document could not be retrieved. A younger clock estimate does not establish durable rejuvenation or bioelectric causation.
Read the full evidence record or inspect the published aggregate fixture. The fixture transcribes reported summaries; it does not invent underlying samples.
Ask whether a changed state preserves appropriate identity and improves the cell’s job.
Separate geometry, contact, force and coupling from altered cell composition.
Distinguish coverage and regrowth from durable, integrated function.
Audit donors, clock provenance, functional outcomes and alternative explanations.
Import a local CSV or edit a published aggregate fixture. Keep molecular, functional, identity and safety evidence side by side.
Compare passive conductance networks with reproducible, synthetic presets and downloadable voltage time courses.
Extend the existing brief with donor replication, matched controls, blinded analysis, durability and rights questions.
Five original-source records, original publication dates, explicit access limits and next tests.
First audit available data for electrical-state/function relationships and confounders. No Karam experiment is claimed.
Reconcile the Anthrobot clock discrepancy and sample hierarchy before interpreting morphology-associated clock differences.
Later laboratory work requires separate scientific, safety and sourcing review. These briefs are high-level research design, not cell-culture or treatment protocols.
Proposed scopes only. Qualified scientific review, authorized data, usable rights, capacity and written terms must be established before an engagement. No staff, customers, laboratory capacity or prices are asserted here.
Karam investigates functional restoration. Project First Cell studies how a defined seed and environment produce organized behavior. This is a shared question about organization, not evidence that living cells emerged from nonliving chemistry, nor that an Anthrobot experiment tests First Cell’s hypothesis.
Review prior art and rights before developing an implementation. Patentability, ownership, licensing and freedom to operate are separate, unresolved questions. The broad idea of antiaging/pro-regenerative screening is already described in WO2026035859A1; it is not presented as new Karam IP. A patent-database status label does not grant permission.
