Selective Signaling Control

Interrupt the damage. Preserve the maintenance.

Concept illustration / not experimental evidence
The program question

Can a harmful interaction be altered without sacrificing normal lysosomal maintenance or protective growth controls?

Investigate whether an unwanted signaling function can be separated from a protein’s essential cellular work. GNPTAB–STING is a documented external lead, not a Karam-owned or validated target.

01 / WORKSTREAM

Map the functions separately

Distinguish the reported noncanonical signaling interaction from the protein’s maintenance roles; preserve cell-type and perturbation context.

02 / WORKSTREAM

Define independent confirmation

A proposed follow-up should test specialized function, cell identity and alternative explanations rather than relying on one attractive senescence marker.

03 / WORKSTREAM

Consider more than editing

A credible target might ultimately support a drug, RNA, protein or other modality. CRISPR can be a discovery method without being the eventual medicine.

What would justify the next step?

A selective effect must be reproduced and explained before a development asset is claimed. Rights, safety and target-specific delivery remain separate workstreams.

From question to decision

A program names the scientific direction. A project brief defines a proposed bounded work package. A commercial scope specifies what a buyer would receive and what must be qualified before contracting. These are related, but they are not interchangeable.

Research context

External work. Explicit limits.

GNPTAB–STING: separate harmful signaling from maintenance

A focused Cas9 screen identified GNPTAB as a regulator of senescence. The assessment describes a noncanonical interaction with STING and a binding-deficient mutant that retained the tested lysosomal functions without inducing senescence.

Target / subject
GNPTAB / STING
Direction
Disruption; overexpression; interaction separation
Cell / setting
Human mesenchymal progenitor cells
Boundary
This is mechanistic human-cell evidence, not treatment of people. Deleting or suppressing GNPTAB everywhere would not follow from an interaction-specific result.

Off-target findings differ across cells and organs

Single-cell and in vivo profiling found heterogeneous off-target and translocation outcomes, including events obscured by bulk analysis. Some experiments deliberately used a promiscuous guide.

Target / subject
Guide- and tissue-specific loci
Direction
Off-target / translocation measurement
Cell / setting
Individual cells and mouse tissues
Boundary
The work does not provide a universal error rate for every editor or clinical guide. Results depend on the guide, cell population, delivery and assay sensitivity.

POLR2A activation: cellular findings are not a checkpoint bypass license

Activating endogenous POLR2A with CRISPRa alleviated senescent phenotypes in the reported system. The assessment describes a pathway connecting LMO7-mediated regulation with MDM4, p53 and p21.

Target / subject
LMO7 / POLR2A / MDM4-p53-p21
Direction
Activation
Cell / setting
Senescence-associated cellular model
Boundary
This does not show that broad suppression of protective p53 signaling is a healthy-aging strategy. Specific context, identity and growth safeguards must remain visible.
